The following is from a post on another board. The poster is a graduate of Harvard Medical School and Fellowship at MIT. He was releaved of his medical license because he prescribed AAS to a patient. He is an advocate for the responsible use of AAS.

The content is related to a question on the board about Tren and gyno. This seems to contradict conventional wisdom about Tren. Everyone raves about how it causes gyno through the expression of prolactin.

I went to the trouble of writing one of the foremost experts on trenbolone. He provided with me with a wealth of information on trenbolone. I asked about trenbolone gynecomastia. Both he and I agree that it is not described in the literature.] Further, any association would be worthy of publication.

Here are his thoughts on the interactions of trenbolone with the progesterone receptor (PR). The literature indicates that 17beta-trenbolone has a high affinity for the bovine PR (Bauer ER et al 2000); while its primary metabolites 17alpha-trenbolone and trendione do not (~1-2% of the affinity of progesterone). The ED50 (effective dose) of trenbolone to produce activity at the PR in a yeast bioassay (40nM) is approximately 10-fold higher than progesterone (4.5nM) and 57-fold higher than THG (0.7nM), a highly progestogenic AAS (Death AK et al 2004). Similarly, others have shown that trenbolone has a high ED50, with a relative potency at the PR near that of testosterone (i.e., almost non-existent) using yeast bioassays (McRobb L et al 2008). Together, these results appear to suggest that trenbolone does in fact bind to the PR, but that it requires a relatively high dose to induce biologic activity (compared with progesterone or THG for example). This would seem to agree with previous reports that trenbolone is either non-progestogenic or only weakly progestogenic (Neumann F 1976).

As a side note, I would invite you to write a Letter to the Editor of the journal Steroids, in response to our recently published report, regarding your clinical experiences with trenbolone induced gynecomastia. I believe this would be of great interest to other clinicians, especially considering the preponderance of evidence suggesting that trenbolone is only a weak progestin and only weakly estrogenic (see our review for discussion).

i firmly believe that tren does not cause gyno, the problem is
1 tren is faked too much so lots of guys end up using a different substance
2 guys use tren in a stack usually with more than one other aa
3 there is no possible way of knowing what is causing the gyno, i mean is it prolactin, progesterone, or estrogen?
4 tren's molecule does not share any similarity with progesterone's

Well prolactin itself does not cause gyno. It is the hormone responsible for milk production in pregnancy. tren most likely will not cause gyno. I have never said it did. It will stimulate prolactin activity, or I should rephrase and say it seems to invoke prolactin thus weeping nipples in men who use it. I agree that tren itself most likely will not give you gyno, oestrogen does. Or aromatization of test into oestrogen. Goos bit of research. Just remember, tren was never intended for human use. It was invented to bulk up beef cattle very quickly. ;)

Great article cosmic! I would also have to agree that I don't believe that tren causes gyno. In my experience with it, tren doesn't seem to be a culprit in any account for I have never gotten it. It doesn't really seem very strong either in progesterone effects from my experience.

Please read my post again.....I never said it was not used by humans, it was never intended for use by humans and as such the FDA has never approved the use of tren for humans. Tren is illegal in the USA except for use in veterinary medicine. It was originally invented in the early 70's for veterinary use in cattle. Underground labs started making tren and derivatives for human use.
